Abstract
The paramount objective in this experiment is to isolate intact RNA to study the signaling pathway involved in the induction of defense related proteins in the cells of Dioscorea bulbifera (air potato). Another inquiry is the role of salicylate as an activator of the defense response in air potato, even in the absence of bacterial and fungal elicitors. The first and most important step in this experiment is to isolate clean, intact RNA to assess the gene expression of defense- related proteins. Standard RNA isolation methods do not yield usable RNA, putatively due to the presence of phenolic compounds. Here, we test modifications to standard RNA isolation procedures to yield adequate RNA sufficient to ultimately identify components of the defense response of air potato.
